Semax

Nootropic / BDNF Upregulator

Also known as: MEHFPGP · Synacthen analog

A synthetic heptapeptide ACTH analog developed in Russia with cognitive-enhancing, neuroprotective, & BDNF-upregulating properties. Approved for stroke & cognitive deficit treatment.

Typical Dose

300–600 mcg 1–2x daily

Route

Intranasal (most common) or subcutaneous injection

Cycle

2–4 weeks on, 1–2 weeks off

Half-life

~7 minutes (active peptide); BDNF effects persist for days

Storage

Reconstituted: 2–8°C, use within 30 days.

Overview

Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) derived from the adrenocorticotropic hormone (ACTH) fragment ACTH(4-10) with added Pro-Gly-Pro for stability. It is approved by the Russian Ministry of Health for treatment of stroke, memory disturbances, & attention deficit disorders.

Its primary mechanisms include upregulation of BDNF (brain-derived neurotrophic factor) & NGF (nerve growth factor) synthesis, potentiation of dopaminergic & serotonergic neurotransmission, & neuroprotective effects against ischemia-reperfusion injury. Unlike stimulants, its cognitive enhancement is mediated through neuroplasticity rather than neurotransmitter depletion.

Quick Start Guide

1

Reconstitute with 1 mL bacteriostatic water per 5–10 mg vial.

2

Intranasal: 300–600 mcg (3–6 drops per nostril) once or twice daily.

3

SubQ: 300 mcg once or twice daily.

Research Indications

Cognitive enhancement

Most Effective

Russian approved for cognitive deficit. BDNF upregulation, improved memory consolidation, attention, & working memory in controlled studies.

Stroke recovery / neuroprotection

Most Effective

Approved for ischemic stroke treatment in Russia. Reduces neurological deficit in post-stroke patients. Neuroprotective mechanism via BDNF & NGF.

Anxiety & depression

Moderate

Serotonergic & dopaminergic modulation produces mood-stabilizing effects. Less potent than Selank for pure anxiety.

Peptide Interactions

SelankSynergistic

The classic nootropic combo: Semax (cognitive stimulation, BDNF) + Selank (anxiolytic, focus). Often taken together at separate times of day.

Side Effects & Safety

Common

  • Nasal irritation (intranasal)
  • Mild stimulation or restlessness at higher doses
  • Vivid dreams

Uncommon

  • Headache
  • Irritability

When to Stop

  • Signs of allergic reaction
  • Active seizure disorder (theoretical concern)

How to Reconstitute

1

Reconstitute with 1 mL BAC water per 5–10 mg vial.

2

Transfer to nasal spray for intranasal use.

3

Refrigerate.

Dosing math: 1 mL BAC water per 5 mg = 5000 mcg/mL. For 300 mcg: 0.06 mL.

Quality Indicators

Good — use as normal

  • Clear, colorless solution

Acceptable

  • Very slight tint

Discard immediately

  • Cloudiness
  • Particulate

What to Expect

Day 1–5

Mental clarity & focus improve noticeably within the first few doses. Stimulating effect is mild.

Week 2–4

BDNF upregulation accumulating. Memory & learning improvements more pronounced.
